The Genomics Core Facility offers the use of two Agilent 2100 Bioanalyzers to assist
investigators in evaluation of their RNA, DNA, and protein samples.
The
Agilent 2100 Bioanalyzer
is a microfluidics-based platform that is used for the quantification and
qualification analysis of DNA, RNA, proteins, and cells. Agilent’s 2100 Expert software
provides quality and quantity digital data for up to 12 samples in 30 minutes. The
results are shown in a gel-like image, electropherogram and tabular formats.
The Genomics Core provides all components of the RNA 6000 Nano kit. Contact
GCF
well in advance of your scheduled run to obtain reagents.
Other kits that include
sample-specific reagents and chips must be ordered by the researcher for
use on the bioanalyzer.

An appointment must be scheduled for each chip run on the Agilent.
Run time on the instrument is 30 minutes. Allow approximately one hour
to set up your chip prior to your scheduled instrument run.
Both
bioanalyzers may be run at the same time. It is recommended to stagger
appointments for each instrument by 15 minutes to avoid setup conflicts or delays.
Appointments may be reserved by Pennington employees using the Outlook calendar.

RNA Analysis
Integrity checks and sample quantitation are essential for doing any RNA application.
The
RNA 6000 Nano and RNA 6000 Pico kits
are used to quantify RNA
using 1 µL of a RNA sample. The 2100 bioanalyzer expert software generates the
sample concentration, the RNA Integrity Number (RIN), and will calculate
ribosomal ratios of the total RNA sample. The bioanalyzer software generates
an electropherogram and a gel-like image for each sample. The electropherogram
provides a detailed visual assessment of the quality of an RNA sample, whereas
RIN can be used as a standard integrity measure that is independent of
concentration, instrument or analyst.
When working with RNA, always keep the area RNase-free; use RNase-free reagents
and chips to avoid sample degradation during analysis.
DNA Analysis
The
DNA 1000, DNA 7500 and DNA 12000 kits
allow you to automatically size and quantitate PCR fragments
and restriction digests accurately and reproducibly. Only 1 µL of
material is required per analysis. The bioanalyzer gives automated
quantitation of each DNA fragment that is normalized against two
internal markers and a ladder for sizing accuracy and reproducibility.
Its broad linear dynamic range enables the detection of less abundant
products and is sensitive enough for the detection of fragments down to 0.1 ng.
Protein Analysis
The
Protein 80 and Protein 230 kits
are capable of providing size, purity and concentration information for ten different
protein samples. The Protein kits are capable of analyzing a large variety of protein samples:
cell lysates, column fractions, antibodies and purified proteins. Only 4 µl of protein sample
is required per analysis. The 2100 Expert software allows for both relative and absolute
quantitation in one assay.
